    Depending on the quality and detail of the glass negative I would probably opt to scan at 1200 or 2000 ppi. Scanning a 2 1/4 “ negative at 2400 ppi gives file size of 2.25” x 1200ppi = 2700px. Taking the long and short sides into account, in this case the calculation is 2700x 2700px = 7,290,000px or a 7.29mb file.

    In this article an Epson V700 scanner is used to scan glass plates. This high-quality flatbed scanner has an optical resolution of 4800 dpi for reflective media (photos, documents) and 6400 dpi for transmissive media (film, glass). The optical density is 4.0 Dmax. Epson now has a successor with the V800, but the V700 is still a very good scanner.

    Emulsion down, with an index card at each end to space it off the glass. This improves sharpness, since these Epson scanners focus about 0.5mm above the glass. I scanned at 1200dpi, rather than the full optical resolution of 2400 dpi, since I wanted the scans to take less than an hour each. (Newer Epson scanners have much better throughput.)
